Cell-free DNA (cfDNA) in blood is widely used as a clinical biomarker. However, the main technical challenges limiting the clinical application of cfDNA are genomic DNA (gDNA) interference, cfDNA degradation, crosslinking reactions, and the preservation of cell activity. Therefore, the ORSIN tube effectively addresses these issues.
This product is used for venous blood storage and transportation, as well as for stabilizing subsequent cfDNA detection. It is a vacuum glass/PET blood sample collection tube. The product is suitable for non-invasive prenatal screening, tumor early diagnosis, efficacy evaluation, and related research.

Product Effectiveness

The performance of the ORSIN tube was compared with that of EDTA, Competitor Tube 1, and Competitor Tube 2. The evaluation indicators included: 1) preventing the release of cellular genomic DNA; 2) inhibiting cfDNA degradation and maintaining cfDNA stability; 3) crosslinking reactions between nucleic acids and other biomolecules; and 4) maintaining cell activity for compatibility with cell nucleic acid tests. The main evaluation methods were: quantitative analysis of different genes and 2100 testing for gDNA release; evaluation of cfDNA degradation using exogenous synthetic DNA sequences and endogenous gene quantification; and judgment of crosslinking reactions based on 2100 peak deviations.Static and transportation tests showed that the ORSIN tube can be stored at ambient temperature for 14 days and transported for 7 days, effectively preventing gDNA release and maintaining cfDNA stability without degradation. It also prevents hemolysis. Compared with similar products, the ORSIN tube has significant advantages.

1 Static Test

The release of genomic DNA (gDNA) from nucleated cells and cfDNA degradation were independently evaluated to avoid interference between these two indicators. EGFR-L858R (Figure 1A) and Kras-G12D (Figure 1B) wild types indicate gDNA release. With prolonged static time, Competitor Tube 1 showed a significant increase in Cp values after 3 days of static storage compared to the ORSIN tube. Exogenous EGFR-L858R (Figure 1C) and Kras-G12D (Figure 1D) mutations indicate nucleic acid degradation. Test results showed that Competitor Tube 1 had more degradation than the ORSIN tube. Within 14 days of static storage, the ORSIN tube was significantly effective in preventing gDNA release from nucleated cells and cfDNA degradation.

Two DNA fragments containing EGFR L858R and Kras G12D mutations (150bp) were synthesized and added to blood collection tubes (one tube per time point for the same healthy individual). After 14 days of static storage at 25°C, plasma was separated, and cfDNA was extracted from plasma (QIAamp® Circulating Nucleic Acid) for qPCR detection.
